Après que son Centre pour les enfants analphabètes s'écrase sur sa femme Matilda et que les services sociaux lui enlèvent la garde de son fils, Derek Zoolander se retire dans un chalet loin des regards, espérant y finir sa vie. Mais un jour, Billy Zane lui apporte un message : un des plus grands stylistes du monde veut qu'il défile pour lui. Zoolander se rend donc à Rome et rencontre Hansel en chemin, qui a reçu la même invitation. Là-bas, ils sont abordés par Valentina, qui leur apprend que des popstars sont éliminées par un mystérieux assassin.
